چکیده

In recent years, research in the field of stem cells is rapidly expanding and the published data are of great promise for the future of the regenerative medicine. Mesenchymal stem cells (MSC) are a relatively new direction in these studies. Their ability to differentiate to a number of cell types indicates that they have the potential to be used as therapy for a variety of medical conditions. Along with the aforementioned, in general public a discussion was forwarded regarding the moral standards in science and the use of human embryos and foetuses for stem cells research. Therefore, our investigations are aimed at isolation and culture of the adherent fraction of ovine fetal liver cells and their introduction as an experimental in vitro model. Our results showed that the isolation of the cells is comparatively easy and that they are capable of long-term culture without differentiation. In addition, the adherent ovine fetal liver-derived cells have high proliferative capacity and exhibited similar morphological characteristics to MSC. According to these findings we suggest, they could be successfully used instead of human material in fundamental research, in order to avoid ethical restrictions.

Differentiation Potential and Culture Requirements of Mesenchymal Stem Cells from Ovine Bone Marrow for Tissue Regeneration Applications

Objectives- To isolate, culture-expand and differentiate mesenchymal stem cells from ovine bone marrow and determine their culture requirements for high expansion rate. Design- Experimental study. Animals- Five Shal sheep. Procedures- In this study, ovine marrow cells were plated and culture expanded through 3 successive subcultures. The resultant cells were then plated at differentiating condi...

Expansion of Non-Enriched Cord Blood Stem/Progenitor Cells CD34+ CD38- Using Liver Cells

Many investigators have used xenogeneic, especially murine stromal cells and fetal calf serum to maintain and expand human stem cells. The proliferation and expansion of human hematopoietic stem cells in ex vivo culture were examined with the goal of generating a suitable protocol for expanding hematopoietic stem cells for patient transplantation. Using primary fetal liver cells, we established...

Increasing fetal ovine number per gestation alters fetal plasma clinical chemistry values

Intrauterine growth restriction (IUGR) is interconnected with developmental programming of lifelong pathophysiology. IUGR is seen in human multifetal pregnancies, with stepwise rises in fetal numbers interfering with placental nutrient delivery. It remains unknown whether fetal blood analyses would reflect fetal nutrition, liver, and excretory function in the last trimester of human or ovine IU...

Histological study of the atrioventricular node and bundle in the heart of ovine fetus

This study was conducted on the atrioventricular node (AVN) and atrioventricular bundle (AVB) of fivefour-month-old ovine fetuses. The histological structure of these components was studied by routinehistological techniques and use of specific staining methods. The AVN was caudally located adjacent to theroot of the aorta. It was almost spherical in shape and consisted of twisty cells. The node...

Early oleate deficiency leads to severe defects in fetal rat liver development

Objective(s): Oleate can be produced through de novo synthesis, which contributes to biological processes and signaling pathways. However, the role of this non-essential fatty acid in hepatic development remains unclear. The current study aimed to evaluate the influence of early oleate deficiency induced by the inhibitor of de novo oleate synthesis MF-438 on fetal rat ...
